Orf virus (ORFV), a member of parapoxvirus, is an enveloped virus with genome of double-stranded DNA. ORFV causes contagious pustular dermatitis or contagious ecthyma in sheep and goats worldwide. In general, detection of viral DNA and observing ORFV virion in tissues of afflicted animals are two methods commonly used for diagnosis of orf infection; however, isolation of the ORFV in cell culture using virus-containing tissue as inoculum is known to be difficult. In this work, the ORFV (Hoping strain) isolated in central Taiwan was successfully grown in cell culture. We further examined the biochemical characteristic of our isolate, including viral genotyping, viral mRNA and protein expression. By electron microscopy, one unique form of viral particle from ORFV infected cellular lysate was demonstrated in the negative-stained field. Moreover, immunomodulating and anti-influenza virus properties of this ORFV were investigated. ORFV stimulated human monocytes (THP-1) secreting proinflammatory cytokines IL-8 and TNF-α. And, pre-treatment of ORFV-infected cell medium prevents A549 cells from subsequent type A influenza virus (IAV) infection. Similarly, mice infected with ORFV via both intramuscular and subcutaneous routes at two days prior to IAV infection significantly decreased the replication of IAV. In summary, the results of a current study indicated our Hoping strain harbors the immune modulator property; with such a bio-adjuvanticity, we further proved that pre-exposure of ORFV protects animals from subsequent IAV infection.  相似文献

Studies that investigate the underlying mechanisms of disease and treatment options typically require the use of a suitable animal model. Few suitable animal models exist for left atrial thrombosis. Here, we demonstrated that the Spontaneously-Running-Tokushima-Shikoku (SPORTS) rat — a Wistar strain known for its running ability—is predisposed to the development of thrombi in the left atrium. We investigated the incidence of left atrial thrombosis in male (n = 16) and female (n = 17) SPORTS rats and observed organized atrial thrombosis in 57% and 38% of males and female rats, respectively. In the male rats, systolic blood pressures and heart rates were significantly higher in SPORTS rats than in control Wistar rats. We could not find any evidence of arrhythmias, such as atrial fibrillation, during electrocardiographic examination of SPORTS rats. We believe that the SPORTS rat could serve as a new research model for left atrial thrombosis; further, it may be suitable for research investigating the development of new antithrombotic approaches for the control of atrial thrombosis or familial thrombophilia in humans.  相似文献

The objective of this study was to examine the health and meat production of cloned sows and their progenies in order to demonstrate the application of somatic cell cloning to the pig industry. This study compared the growth, reproductive performance, carcass characteristics and meat quality of Landrace cloned sows, F1 progenies and F2 progenies. We measured their body weight, growth rate and feed conversion and performed a pathological analysis of their anatomy to detect abnormalities. Three of the five cloned pigs were used for a growth test. Cloned pigs grew normally and had characteristics similar to those of the control purebred Landrace pigs. Two cloned gilts were bred with a Landrace boar and used for a progeny test. F1 progenies had characteristics similar to those of the controls. Two of the F1 progeny gilts were bred with a Duroc or Large White boar and used for the progeny test. F2 progenies grew normally. There were no biological differences in growth, carcass characteristics and amino acid composition among cloned sows, F1 progenies, F2 progenies and conventional pigs. The cloned sows and F1 progenies showed normal reproductive performance. No specific abnormalities were observed by pathological analysis, with the exception of periarteritis in the F1 progenies. All pigs had a normal karyotype. These results demonstrate that cloned female pigs and their progenies have similar growth, reproductive performance and carcass quality characteristics and that somatic cell cloning could be a useful technique for conserving superior pig breeds in conventional meat production.  相似文献

ABSTRACT:     The biomass of bighand thornyhead Sebastolobus macrochir was increased by the high recruitment success of the 1999–2002 year classes off the Pacific coast of northern Honshu, Japan. In this study, the growth of bighand thornyhead was examined over a 9-year period from 1996 to 2004 in this area. The growth of the 1999 year class and the 2000–2002 year classes was reduced at 3 and 2 years old, respectively, while the 1999–2002 year classes were smaller than the 1993–1998 year classes. In 2-, 3- and 4-year-old fish, the relationship between abundance and mean standard length was expressed by negative linear regressions, while fish became smaller when abundance of the year class was larger. Mean bottom temperatures were stable at depths of 350–900 m; variations in water temperature were small in the main distribution area of bighand thornyhead. We discuss the factors affecting the growth of bighand thornyhead via changes in the demersal fish community and feeding habits.  相似文献

ABSTRACT: The complete nucleotide sequence of the mitochondrial genome for the Japanese anchovy Engraulis japonicus (Teleostei: Clupeiformes) was determined. The entire genome was purified by gene amplification using the long polymerase chain reaction (PCR) technique, and products were subsequently used as templates for PCR with 56 fish-versatile primers that amplify contiguous, overlapping segments of the entire genome. Direct sequencing of the PCR products demonstrated that the genome (16 675 base pairs [bp]) contained the same 37 mitochondrial genes (two ribosomal RNA, 22 transfer RNA and 13 protein-coding genes) as those found in other vertebrates, with the gene order being identical to that in typical vertebrates. A major non-coding region between the tRNAPro and tRNAPhe genes (1024 bp) was considered to be the control (D-loop) region, as it has several conservative blocks characteristic to this region.  相似文献

The siderophore avaroferrin (1), an inhibitor of Vibrio swarming that was recently identified in Shewanella algae B516, was produced by heterologous expression of the biosynthetic gene cluster cloned from a deep-sea sediment metagenomic DNA, together with two analogues, bisucaberin (2) and putrebactin (3). Avaroferrin (1) is a macrocyclic heterodimer of N-hydroxy-N-succinyl cadaverine (4) and N-hydroxy-N-succinyl-putrescine (5), whereas analogues 2 and 3 are homodimers of 4 and 5, respectively. Heterologous expression of two other related genes from culturable marine bacteria resulted in production of compounds 1–3, but in quite different proportions compared with production through expression of the metagenomic DNA.  相似文献

ABSTRACT:   High- and multi-frequency acoustic measurement systems and the multi-frequency inversion (MFI) method have been used to measure spatial distributions and abundances of zooplankton by size. In this study, the calibration method for high- and multi-frequency systems was developed and the validation of MFI method was carried out by scatterer measurement. The standard sphere calibration method that has not been applied to such high- and multi-frequencies was applied to calibrate our high- and multi-frequency system, TAPS-6 (Tracor Acoustic Profiling System, BAE Systems). An optimum size of standard sphere of tungsten carbide of 1 mm radius was derived to have a small target strength variation for the six frequencies of TAPS-6, and the practicability and precision of the standard sphere calibration method was confirmed for those frequencies. A school or cluster of dummy scatterers of zooplankton with small tungsten carbide spheres were designed to validate the MFI method, and volume back-scattering strength values were measured by the multi-frequency system. By comparing the result of the inversion with their real composition, the features of the MFI method could be validated and examined.  相似文献

Leaf-lamina resistance, R(L), accounts for a large fraction of branch resistance across a wide range of plant species. This work hypothesized that large R(L) is essential for distributing water equally to leaves on the shoot, and tested this hypothesis through theoretical analyses and measurements using over 10-m-long current-year shoots of kudzu vine, Pueraria lobata [Willd.] Ohwi. First, the hydraulic architecture and the distribution of the motive force achieving equal distribution of water delivery were theoretically obtained by simulating water flow through a hypothetical shoot comprising an axial pathway and several lateral pathways as a stem and leaves, respectively, in a kudzu-vine shoot. The model predicts that large resistance of the lateral pathway relative to that of the axial pathway is associated strongly with small variation in the hydraulic conductance of a pathway from the base of the axial pathways to the lateral pathway among the nodes, rendering water delivery to each lateral pathway equal under small variation in motive force for water flow. For the kudzu-vine shoot, the measured ratio of the lateral (a petiole) to the axial (a stem) resistance was 115. When R(L) was added to the lateral pathway, the ratio increased to 1136. According to the model prediction, these values imply that the hydraulic conductance of a pathway comprising a stem and a petiole, K(BP), is favored strongly at the basal nodes, while the hydraulic conductance of a pathway including a stem, a petiole and a lamina, K(SL), is slightly different across the nodes. For the shoots with leaf lamina, the diurnal change in transpiration rate was not different between the leaves on the three nodes dividing the shoot into four parts. K(SL) was not related significantly to node number. Conversely, K(BP) at the distal node was ~0.06-fold that at the basal node. Furthermore, the motive force for water flow should vary by 6.64-fold among nodes to compensate for the favored distribution of K(BP), which is an unrealistic value. These results indicate that R(L) contributes largely to an equal distribution of water delivery in a shoot, supporting our hypothesis.  相似文献
